Abstrak

Sistem dispersi padat adalah dispersi satu atau lebih bahan aktif pada suatu matriks atau pembawa di kondisi padatan. Dispersi padat mengarah ke kondisi padat suatu zat yang mengalami dispersi pada material lainnya. Daripada formula yang biasanya digunakan, dispersi padat bisa disusun menggunakan bermacam cara dan mempunyai berbagai kelebihan. Dispersi padat merupakan gabungan obat hidrofobik dan matriks hidrofilik. Obat pada dispersi padatan bisa terdispersi melalui bentuk molekul, padatan amorf, maupun kristal. Pembawa yang digunakan dalam sistem dispersi padatan yakni polimer. Saat polimer serta obat terjadi interaksi maka obat mengisi rongga diantara rantai-rantai polimer dan menyebabkan rantai polimer cenderung fleksibel. Rendahnya kelarutan bahan aktif dalam air menyebabkan rendahnya laju disolusi dan bioavailabilitas sehingga mempengaruhi tingkat keefektivan terapi. Maka dari itu, solubilitas adalah tolak ukur krusial ketika merancang sediaan yang berkualitas, aman, serta efektif. Metode dispersi padat telah banyak dimanfaatkan guna mengoptimalkan solubilitas serta disolusi bahan aktif obat. Solubilitas serta laju disolusi dapat ditingkatkan melalui bermacam mode pembentukkan dispersi padat diantaranya Dropping Method, Spray Drying Co-grinding, Lyophilization Technique, Fusion Method, Kneading, and Solvent Evaporation. Bermacam jenis polimer pembawa yang bisa dimanfaatkan untuk membuat dispersi padatan juga dibahas pada artikel ini. Hasil review artikel memperlihatkan bahwasanya kelarutan solubilitas serta laju disolusi bahan aktif obat dapat ditingkatkan menggunakan metode dispersi padat yang disandingkan dengan bahan aktif murni.
