Formulation and Antioxidant Activity Test of a Moisturizing Gel Preparation from Celery Leaf Extract (Apium graveolens L.)

Abstract
Background: Free radicals are one of the causes of skin problems and premature aging, so antioxidants are needed to inhibit this process. Celery leaves (Apium graveolens L.) are known to contain flavonoids, tannins, saponins, and steroids that have antioxidant potential. Objective: To formulate celery leaf extract into a moisturizing gel, evaluate the physical properties of the formulation, and test its antioxidant activity. Methods: This study is a quantitative experimental study involving maceration extraction with 70% ethanol, formulation of the product, evaluation of physical properties, and antioxidant testing using the DPPH method. Results: The results showed that the celery leaf extract was successfully formulated into a moisturizing gel that met the physical requirements for gel formulations, as indicated by good organoleptic properties and homogeneity, a pH within the skin’s pH range, spreadability and adhesion meeting standards, viscosity within the required range, and stability without phase separation. The IC50 value for vitamin C of 7,4 µg/mL is classified as very strong, whereas the IC50 values for formulations F0, F1, F2, and F3 were, respectively, 3968,6 µg/mL, 7607,1 µg/mL, 4179,3 µg/mL, and 3060,2 µg/mL. Based on these IC₅₀ values, the antioxidant activity of the celery leaf extract moisturizing gel formulations is classified as weak (>200 µg/mL); however, an increase in extract concentration shows a trend toward increased antioxidant activity. Conclusion: This study demonstrates that celery leaf extract was successfully formulated into a moisturizing gel with good physical properties; however, further optimization of the formulation is needed to enhance its antioxidant activity so that it can provide optimal benefits as a skin care product.
