Abstract

Background: Rice quality assessment requires analytical evidence to distinguish potentially chlorine-reactive residues from ordinary physical characteristics. Objective: To examine colorimetric responses and estimate chlorine-equivalent levels in rice sold at three markets in Medan. Methods: Ten white-rice samples were selected purposively from Simpang Limun, Sukaramai, and Petisah markets. Aqueous extracts were examined using N,N-diethyl-p-phenylenediamine (DPD), potassium iodide–starch, silver nitrate, and povidone-iodine reagents. Five DPD-reactive samples were analysed by visible-range spectrophotometry at 510 nm, with six instrumental readings per sample. Results: Samples A–E (50%) produced pink and blue responses in the DPD and iodide–starch tests, respectively; samples F–J remained colourless in both tests. Calibration at nominal concentrations of 0–60 µg/mL yielded R² = 0.9972. Conditional semiquantitative estimates for A–E were 182.99, 220.57, 69.72, 75.12, and 84.00 µg/g, espectively. All samples were white and had a characteristic rice odour. Conclusion: Five samples exhibited oxidant responses consistent with active chlorine. Incomplete matrix validation and an unverified mass-conversion factor limit quantitative interpretation and preclude definitive conclusions about chlorine content or food-safety compliance.
